Which barrier still blocks the cell-state change?

A target list answers only part of the question

Ranking targets individually can identify promising interventions. A staged model asks whether the proposed set can get a cell population through the required sequence of recruitment and differentiation. It distinguishes a necessary step from an intervention that merely scores well on its own.

A concrete remyelination question

The remyelination analysis asks whether downstream differentiation drive can bypass an inhibitory constraint. If the barrier remains under that test, a differentiation-only proposal is insufficient in the model. That is a useful reason to change the combination or the experiment.

Use the right biological setting

Lesion classes, cell populations and non-monotonic responses matter. Current work supports constrained necessity reasoning, not a quantitative prediction of remyelination. Flow-based calculations require suitable measured capacities. Removing a necessary barrier does not by itself establish that repair will follow.

From a barrier to a combination hypothesis

Resistance modeling can supply a complementary question about escape routes; spatial information may eventually identify where a barrier operates. BioTwin’s full spatial integration remains a research direction.
